An experimental Study on Inoculation Method of Typhoid Vaccine and Immunological Responses by Macrophages


Abstract
It was a subject to determine whether typhoid immunity is mainly correlated vitir antibody production or with enhanced activity of phagocytes.
To investigate this problem, Salmonella t phi Ty-2 and 0901 were employed. After immunization of Salmonella typhi(Ty-2 or 0901) in rabbit to three intr dermal or subcutaneous injection, agglutinin titer and phagocytic rate were compared. In order to study immunological responses by peritoneal macrophages four combination as immune macrophages suspension in cell maintenance fluid enriched Faith immune serum, normal macrophage suspension in cell maintenance fluid enriched. with immune serum, immune macrophages suspension in cell maintenance fluid. enriched with normal serum and normal macrophages suspension in cell maintenance fluid enriched with normal serum were prepared from immune serum containing various agglutinin titer, peritoneal macrophages from immune rabbit above various agglutinin titer, normal rabbit serum and peritoneal macrophages with from normal rabbit.
Results were as follows.
1. No significant differences were noted in agglutinin titer between intradermal and subcutaneous inoculation of typhoid vaccine.
2. The phagocytic activities of peritoneal macrophages induced from subcutaneously injected rabbit were more enhanced than from intradermally injected rabbit.
3. There was a tendency toward parallelism in the appearance of agglutinin titer and the phagocytic rate according to degree of agglutinin titer. But in a certain case, the titer of the agglutinin did not necessarily parallel the phagocytic rate.
4. In experimental study on rabbit typhoid immunity, phagocytic rate followed the order of immune macrophages suspension in cell maintenance fluid enriched with.
immune serum, normal macrophages suspension in cell maintenance fluid enriched with immune serum, immune macrophages suspension in cell maintenance fluid
enriched with normal serum and normal macrophages suspension in cell maintenance fluid with enriched normal serum.
5. Typhoid immunity studied by phagocytic rate was correlated with both immune macrophages and immune serum.
